Nucleic acids or proteins in a complex sample can be visualized using electrophoresis and blotting. Gel electrophoresis separates molecules based on charge and/or molecular weight. Once resolved, molecules can be visualized directly on the gel, for example using CyDye fluorescent dyes to label proteins or intercalating ethidium bromide to allow UV detection of nucleic acids.
Blotting involves the transfer of separated nucleic acids or proteins onto a membrane and then detection of the target molecule using labeled antibodies (Western blotting) or labeled nucleic acid probes that recognize complementary DNA (Southern blotting) or RNA (Northern blotting) sequences. Detection systems based on luminescence, fluorescence, or radioactivity can then be used to detect protein or nucleic acid targets, revealing the original migration position, and depending on the detection method, the quantity of the target.
